PolyVitro-D is the economical, low-impact solution to evaluating the release of poorly soluble drugs from drug releasing articles such as coating, microparticles, and other materials. This environmentally responsible polymer based surfactant degrades slowly eliminating any downstream waste issues.
The packet when dissolved in 500 ml of HPLC grade water generates a media that can solubilize hydrophobic drugs for easy analysis.
PolyVitro-D has a simple one-peak UV/Vis absorbance spectrum with relatively little absorbance across a broad spectrum of the visible region.
Test: PLGA (~25kDa (1:1 La:Ga), Boeinger-Ingelheim RG503H) was dissolved in dichloromethane (10% w/v) and cast coated onto 25mm glass circle coverslips and dried in air for 2hours followed by 3 hours under deep vacuum. The formed films were then placed in a 6-well plate and had either Phosphate buffered Saline alone or Phosphate buffered Saline plus 2% polysorbate-20 add to them and were incubated at 37C with 120RPM orbital shaking for a period of 10 days. Afterwards the solutions were removed, the films were rinsed several times with distilled water and then vacuum dried. Subsequently these polymers were redissolved in dichloromethane and tested via GPC using a Varian Prostar system with 1 ml/min DCM flow across two Resipore 3 um columns and one Mixed E column (Varian) and with UV-Vis (PDA)detection. The results were compared to a series of polystyrene calibration standards.
The average molecular weight of PLGA after 10 days in PBS alone was 10419 Da ± 325Da (ttest95% confidence). The average molecular weight of PLGA after 10 days in PBS plus polysorbate-20 was 3756 Da ± 1452Da.
This test shows there is statistically significant difference between PLGA molecular weights after incubation in just PBS as opposed to incubation in PBS + polysorbate-20 indicating increased degradation with polysorbate-20.
